CELL CULTURE MODELS IN PRECLINICAL BIOPHARMACEUTICS
For preclinical biopharmaceutical evaluations, various cell culture models have been established in our group. These comprise the Caco-2 cell line, a nowadays widely accepted model for the human intestinal epithelium, models of prostate-, bladder- and mamma-carcinoma as well as human chondrocyte cell lines and the THP-1 macrophage model. Mainly, these cell culture systems are applied to elucidate preclinical biopharmaceutical aspects of specifically designed drug delivery systems. Among these our main focus is set on the estimation of cytoadhesion and cytoinvasion as well as the intracellular trafficking of drugs and their delivery systems. For a detailed characterisation of these issues, methods including flow cytometry, confocal laser scanning microscopy and tailored fluorescence techniques, particularly in the area of immunofluorescence staining for co-localisation experiments, are applied. Currently, we are developing an acoustically driven mirofluidic cell chip to analyse particle-cell interactions under shear stress. However, our expertise also encompasses traditional horizontal and vertical setups to study transepithelial transport characteristics. Moreover, in collaboration with the Technical University of Vienna microelectronic biosensors coated with viable human cells are being developed. The potential applicability of such devices primarily lies in toxicity screening of drugs and excipients, but additionally might prove to be utilisable for high throughput screening in preclinical drug research and development. Another focus in the field of cell cultures is the detailed characterisation of cell models regarding their differentiation and vitality, especially in view of the impact of surface modifications, and the underlying cell biological mechanisms.
